(-)-epicatechin gallate是绿茶中主要的儿茶素成分[1]。
(-)-epicatechin gallate(ECG)在细胞生长抑制、细胞凋亡和膜转运系统中具有重要作用[1]。
(-)-epicatechin gallate是一种儿茶素。在HCT-116细胞中,ECG激活转录因子3(ATF3),其在促凋亡中发挥关键作用。EGR-1参与ECG诱导的ATF3的表达。在HCT-116细胞中,ECG(50 μM)以时间和剂量依赖的方式增加NAG-1和ATF3的表达。在HSC-2癌细胞中,ECG(50 μM)具有细胞毒性,NR50(midpoint cytotoxicity)值为67 μM。ECG(250 μM)诱导核小体DNA片段化和细胞凋亡。ECG(150 μM)显著增加caspase-3的活性[2]。然而,在正常HGF-2成纤维细胞中,ECG在浓度达到25 μM时才具有细胞毒性,NR50值为100 μM。
在大鼠中,ECG有五种代谢物,分别是(-)-epicatechin gallate、3’,4’’-di-O-methyl-(-)-epicatechin gallate、4’’-O-methyl-(-)-epicatechin gallate、4’-O-methyl-(-)-epicatechin gallate 和3’-O-methyl-(-)-epicatechin gallate,他们均随尿排出[3]。
